本課題組長(zhǎng)期致力于研究組織器官間交流對(duì)話(Inter-organ communication)調(diào)控免疫代謝穩(wěn)態(tài)(Immune-metabolic homeostasis)的時(shí)空交互機(jī)制,探索能量穩(wěn)態(tài)的應(yīng)激響應(yīng)系統(tǒng)調(diào)控肥胖、衰老以及免疫代謝相關(guān)健康穩(wěn)態(tài)的分子機(jī)理,目前已鑒定了肌肉分泌因子、線粒體代謝物及脂滴等能量感應(yīng)因子,揭示了骨骼肌介導(dǎo)各組織器官協(xié)同響應(yīng)感染、晝夜節(jié)律或營(yíng)養(yǎng)應(yīng)激的能量轉(zhuǎn)換機(jī)制,發(fā)現(xiàn)了骨骼肌為核心的免疫代謝響應(yīng)體系,突破了組織器官間的免疫代謝對(duì)話如何適應(yīng)各種應(yīng)激的難題,為研究免疫代謝紊亂相關(guān)的疾病提供了理論基礎(chǔ),共已發(fā)表SCI論文25篇,總計(jì)引用超過(guò)1000次,其中以第一作者發(fā)表SCI論文10篇,文章發(fā)表在Cell Metabolism,Current Biology,Aging Cell,Current Opinion in Insect Science,BMC Genomics等領(lǐng)域內(nèi)權(quán)威或Top期刊;單篇最高引用160多次,h指數(shù)14。相關(guān)研究引起了國(guó)內(nèi)外學(xué)者的廣泛關(guān)注和認(rèn)可,英國(guó)醫(yī)學(xué)科學(xué)院院士Irene Miguel-Aliaga教授為Cell Metabolism文章撰寫亮點(diǎn)評(píng)論文章,另外發(fā)表在Cell Metabolism和Current Biology的文章獲得Faculty Opinions(F1000Prime)推薦。曾主持完成國(guó)家自然科學(xué)基金青年項(xiàng)目(31301951)、中國(guó)博士后面上基金(2013M531638)各一項(xiàng)。目前正在受到西安交通大學(xué) “青年拔尖人才支持計(jì)劃”A類(2022-2028)、陜西省“三秦英才引進(jìn)人才”青年項(xiàng)目和國(guó)家自然科學(xué)基金面上項(xiàng)目(2023-2026)的支持。

研究方向

營(yíng)養(yǎng)及能量代謝的穩(wěn)態(tài)調(diào)控是動(dòng)物應(yīng)對(duì)自然界各種生存挑戰(zhàn)的關(guān)鍵機(jī)制,動(dòng)物需要根據(jù)進(jìn)食狀態(tài)、營(yíng)養(yǎng)水平或病菌感染的動(dòng)態(tài)變化,利用免疫代謝感應(yīng)系統(tǒng)實(shí)時(shí)調(diào)整能量資源的攝取、儲(chǔ)存和利用。因此,能量代謝系統(tǒng)和天然免疫系統(tǒng)之間相互交叉,共進(jìn)化形成了復(fù)雜的免疫代謝交互網(wǎng)絡(luò),在不同營(yíng)養(yǎng)、生理與病理應(yīng)激下調(diào)控能量代謝的適應(yīng)性改變,是維持系統(tǒng)性免疫代謝穩(wěn)態(tài)(immune-metabolic homeostasis)的關(guān)鍵機(jī)制。然而,各種營(yíng)養(yǎng)、飲食、生理與病理應(yīng)激往往誘發(fā)能量代謝紊亂,破壞機(jī)體的免疫代謝穩(wěn)態(tài),導(dǎo)致肥胖、衰老及多種免疫代謝疾病的發(fā)生,如糖尿病、脂肪肝、心血管代謝病、癌癥及感染、炎癥相關(guān)疾病,我國(guó)的人口老齡化則加劇了這些健康問(wèn)題的嚴(yán)峻形勢(shì)。因此,闡明能量的代謝穩(wěn)態(tài)如何響應(yīng)各種應(yīng)激因素的應(yīng)答機(jī)制,對(duì)于研究免疫代謝疾病及健康穩(wěn)態(tài)的調(diào)控機(jī)理,推動(dòng)面向人民生命健康的基礎(chǔ)和應(yīng)用研究具有重要意義。

由于各組織器官具有不同的代謝功能,因此能量代謝的穩(wěn)態(tài)調(diào)控涉及多個(gè)組織器官間的功能互作、代謝模式的時(shí)空協(xié)同和代謝信息的交流對(duì)話。為了研究各組織器官如何協(xié)同響應(yīng)不同能量應(yīng)激的關(guān)鍵科學(xué)問(wèn)題,需要從組織器官間對(duì)話網(wǎng)絡(luò)(inter-organ communication)的維度突破能量感應(yīng)調(diào)控與異常的研究瓶頸。例如,不同營(yíng)養(yǎng)、飲食或生理與病理因素重塑能量代謝的過(guò)程中,骨骼?。芰肯模┡c脂肪(能量?jī)?chǔ)存)、腸及大腦(能量感應(yīng)、消耗及攝取)之間的代謝互作模式和信息對(duì)話機(jī)制仍亟待解決。因此,鑒定不同營(yíng)養(yǎng)、飲食或生理與病理應(yīng)激誘導(dǎo)的代謝酶、代謝物和分泌因子,揭示其介導(dǎo)組織或器官間對(duì)話的信號(hào)通路,闡明能量代謝適應(yīng)性轉(zhuǎn)換的調(diào)控機(jī)制,已成為生命健康研究的關(guān)鍵挑戰(zhàn)。

本課題組聚焦上述面向人民生命健康的關(guān)鍵科學(xué)問(wèn)題,長(zhǎng)期致力于研究器官間對(duì)話調(diào)控免疫代謝穩(wěn)態(tài)的分子機(jī)制,探索各種生理/病理應(yīng)激因素誘發(fā)組織器官間的能量代謝失衡,進(jìn)而導(dǎo)致免疫代謝紊亂并最終影響健康、衰老與壽命的生理機(jī)理。因此,本課題組將系統(tǒng)性闡明器官間能量感應(yīng)體系的適應(yīng)性響應(yīng)機(jī)理,為研究免疫代謝紊亂相關(guān)的疾病和健康穩(wěn)態(tài)提供關(guān)鍵的理論基礎(chǔ),主要研究方向包括:

(1)不同營(yíng)養(yǎng)、生理與病理應(yīng)激下,能量代謝系統(tǒng)與天然免疫系統(tǒng)之間的協(xié)同整合機(jī)制。

(2)組織及器官間的免疫代謝穩(wěn)態(tài)調(diào)控衰老、肥胖、糖尿病、慢性炎癥等免疫代謝障礙的分子機(jī)制。

(3)組織或器官間對(duì)話調(diào)控免疫代謝穩(wěn)態(tài)的應(yīng)激響應(yīng)機(jī)制及其影響健康穩(wěn)態(tài)的分子機(jī)理。
